What problem will arise if the level of developing liquid is higher than the sample spot?Paper Chromatography?
and how important is a good choice of solvent mixture to chromatographic analysis?
and effect/s of having a broad sample shot/ spot on the chromatogram.(all i know is that the results would be difficult to read as they would merge.is that right?)
Im having this pre-laboratory questions on my chemistry lab. I dont really know anything about chromatography. help me pls! thanks!
To answer your first question if your developing liquid is higher than the sample spot you will wash away some if not all of the sample into the developing liquid and as a result ruining your test. To answer your second question, it is incredibly important that you solvent mixture is a good fit for the system if you have an analyte that is very polar you don't want to use a very polar mixture of solvents as it will run you sample spot off the grid the converse would be true if you used a solvent mixture that was completely non polar. Finally, to answer your last question, it will be difficult to determine how many different molecules of varying polarity are in your analyte mixture as the spots will all merge and will just look like a tear dorp a fine drop is ideal as you will be able to clearly see any separations that occur on the paper.
